Cambridge researchers have found an answer to a puzzling aspect of obesity care: drugs with opposite actions on the GIPR receptor can both lead to weight loss, yet they do so through distinct brain regions. GIPR activators suppress appetite via the brainstem, while GIPR blockers work through the hypothalamus to increase feelings of fullness. These conclusions come from experiments in mice, so follow-up studies in humans are essential.
The investigation was carried out by scientists at the University of Cambridge. GIPR-targeting medications come in two opposing varieties: those that activate the receptor and those that block it, and both types reduce body weight. Mounjaro and Zepbound belong to the activator group. MariTide, by contrast, is an experimental drug that blocks the receptor. There are also treatments such as Wegovy and Ozempic that activate a different receptor, GLP-1.
In mice lacking GIPR in specific brain areas, the team found that receptor activation curbed appetite through the brainstem, whereas receptor blockade promoted satiety through the hypothalamus. Blocking GIPR also appeared to enhance the effects of experimental medicines targeting the amylin receptor.
Dr. Jo Lewis, the study's first author, stressed that the results confirm the brain's central role in obesity treatment—rather than pointing only to the gut or pancreas.
She added that the findings are currently based on mouse data alone, making human research a necessary next step. More than one billion people worldwide are living with obesity, a condition that increases the likelihood of type 2 diabetes, cardiovascular disease, and several forms of cancer.
These discoveries may point toward new strategies for developing effective anti-obesity drugs, but the results still need further verification.
